Human Extracellular Matrix

Yleistä

The extracellular matrix (ECM) consists of interstitial connective tissue and basement  membrane (BM). The ECM acts as a backbone for cells and provides a physical barrier. It also influences such functions as cell proliferation, differentiation, adhesion, migration, gene expression, and tissue integrity.

ECM also plays a profound role in tissue injury and healing. The detection of ECM components in various parts of the body provides an efficient tool for following malignant change, invasion and  metastasis. Biohit provides monoclonal antibodies to fi - bronectins(1,2,3), tenascin(4,5), laminin(6,7) and vitronectin.

Monoclonal antibody to
EDA-CELLULAR FIBRONECTIN (cFn) clone DH1 (IgG1)

100 µg immunoglobulin in 1 ml PBS solution containing 1.0% (w/v) BSA and 0.1% (w/v) sodium azide.

Monoclonal antibody (Mab) to cFn is derived from the hybridoma produced by fusion between myeloma cells and Balb/c spleen cells. Purified fibronectin from A8387 fibrosarcoma cells was used as an immunogen (21).

Specificity
The antibody is specific to extradomain A (EDA) sequence of a cellular fibronectin and recognizes thus only the cellular fibronectin. Mab DHI has been shown to specifically block chondrocyte condensation in chicken embryos (5). The antibody crossreacts with human, chicken, rat, rabbit and guinea pig.

Applications
Immunohistochemical stainings, western blotting and other immunoassays for cellular and tumor biology.

Guidelines for dilutions
Working dilution in western blotting is at least 1:1000.


Monoclonal antibody to TENASCIN-C  clone EB2 (IgG1)

100 µg immunoglobulin in 1 ml PBS solution containing 1.0% (w/v) BSA and 0.1% (w/v) sodium azide.

Monoclonal antibody (Mab) to tenascin-C is derived from the hybridoma produced by fusion between myeloma cells and Balb/c spleen cells. Purified tenascin-C from human fetal fibroblasts was used as an immunogen.

Specificity
The antibody reacts with the 4th and 5th fibronectin-like repeats of human tenascin-C polypeptides of Mr 250000 and 180000. The antibody crossreacts with human, guinea pig and rabbit.

Applications
Immunohistochemical stainings, westem blotting and other immunoassays for cellular and tumor biology. Quantitation of Tn-C in body fluids together with Mab DB7 (8).

Guidelines for dilutions
Working dilution in westem blotting is at least 1:1000.


Monoclonal antibody to LAMININ (b1-chain) clone DG10 (IgG1)

C100 µg immunoglobulin in 1 ml PBS solution containing 1.0% (w/v) BSA and 0.1% (w/v) sodium azide.

Monoclonal antibody (Mab) to laminin (b1-chain) is derived from the hybridoma produced by fusion between myeloma cells and Balb/c spleen cells. Partially pepsin-digested laminins from human placenta were used as immunogen (8).

Specificity
The antibody recognizes the Mr 220000 b1-chain of laminin. Mab DG10 appears to react with the NH2-terminus of the Ln b1chain (1).
The antibody reacts with human tissues.

Applications
Immunohistochemical stainings, western blotting and other immunoassays for cellular and tumor biology. Unlike many other Mabs to Ln b1chain Mab DG10 reacts strongly both under reducing and nonreducing conditions. Mab DG10 has been successfully used to purify Ln-8 (2) and Ln-10 (6) by affinity chromatography.

Working dilution in western blotting is at least 1:1000.


Monoclonal antibody to LAMININ (g1-chain) clone BC17 (IgG1)

100 µg immunoglobulin in 1 ml PBS solution containing 1.0% (w/v) BSA and 0.1% (w/v) sodium azide.

Monoclonal antibody (Mab) to laminin g1-chain is derived from the hybridoma produced by fusion between myeloma cells and Balb/c spleen cells. Pepsin-digested laminins from human placenta were used as immunogen.

Specificity
The antibody recognizes the Mr 200,000 g1-chain of human reduced and alkylated laminin produced by various cultured cells, which produce laminins complexed with it. The specificity of Mab BC17 has been confirmed by immunoprecipitation and Elisa by using recombinant g1- chain. The antibody only reacts with human cells and tissues
(further studies are going on).

Applications
Immunohistochemical stainings and immunoprecipitation. Does not work in western blotting.


Monoclonal antibody to VITRONECTIN clone BE10 (IgG1)
